摘要
将免疫组化和原位分子杂交技术应用于流行性出血热尸检石蜡包埋组织,在同一张切片中,进行病毒抗原和核酸的双重检测获得成功。用多种流行性出血热病毒单克隆抗体检测病毒抗原,再用地高华标记的cDNA探针检测病毒RNA,分别采用两种不同的酶显色系统,其结果显示病毒抗原分布于细胞胞浆及小血管基底膜上、而病毒核酸则分布于细胞核。在实验中须注意的是Lugol's液预处理;选择合适的消化酶以及适当的杂交温度和时间。
A combined staning of immunohistochemustry and in sntu hybridization has been successfulty upplied to detect antigen and vira!RNA at the same tissue slce from cpidemic hemorrhagic fever(EHF)autopsy.Multiple McAb were used to detect EHFV antigen.followed with a digoxigenin-labelling cDNA probe for detecting EHFV RNA respectively.The viral anugen was seen in the celtular cytoptasm and viral RNA was demonstrated within the nuctei.It is emphasized that pretreatment of the section with Lugot's solution and choice of suitable proteolytic enzyme as well as proper temperature and time for the hybridization ure the key points for the staining.
