摘要
用ABC免疫组织化学法观察乳酸脱氢酶C_4(LDH-C_4)在性成熟后的Wistar大鼠生精上皮细胞中的定位。结果表明:LDH-C_4定位于粗线期初级精母细胞以后的各级生精细胞中,精原细胞中缺如。显微分光光度分析表明,随着精子的发生,LDH-C_4的相对含量逐渐增加,精母细胞中为0.55±0.09,精子细胞中为0.86±0.13,精子中为0.94±0.04,三者之间有显著性差异(P<0.01)。ABC免疫电镜显示LDH-C_4在大鼠精子的细胞质、线粒体及部分细胞膜上均有分布。免疫荧光显示LDH-G_4在精子表面主要定位于尾部的主段和末段,少量分布于精子头冠部。作者对LDH-C_4细胞定位的生理意义作了讨论。
ABO immunohistochemical staining was used to study the localization of laotate dehydrogenase-C4 (LDH-C4)in the seminiferous epithelia of Wigtar rats. Results indicated that LDH-C4 is localized in the spermatogenio cells of different stages from midphytene sperrftatocybe to mature spm, except spermatogonium. Microspeotrophotometrio determinatioa of the relative enzyme content showed that LDH-C4 increases during spermatogenesis, values were 0.65±0.09 in spermatoeytes, 0.86±0.13 ia gpermatids and 0.94±0.04 in spermatozoa(P<0.01). In the sperm, ABC immunoeleotromicrocope showed thai LDH-C4 is localized in the cytoplasm and mitochondria, and some regions of the sperm membrane.Immunofluorescnet membrane staining showed LDH-C4 to be mainly distributedin the principal and end piece of the sperm tail, and a little in the corona of thesperm head. Its physiological signicance is discussed in relation to the localization of LDH-C4.
