摘要
从仓鼠肝中提取到电泳均一的谷胱甘肽过氧化酶,用微量纯化酶免疫新西兰兔,得到效价为1∶64的抗血清。用该抗血清对仓鼠各组织进行PAP法酶标定位,发现谷胱甘肽过氧化酶广泛存在于肝、肾曲管上皮、心肌、骨骼肌细胞胞浆内。在肝脏、汇管区阳性反应明显多于小叶中央区。
Glutathione peroxidase was successfully purified from hamster liver to give a eingle band in SDS polyaorylamide gel eleotrophoresis. New Zealand rabbits were immunized with microdoses of this enzyme, and a highly specific anti-serum with titer of 1:64 was obtained. Using this antibody, an immunohistoohemical technique (the PAP method,) was applied to investigate the localization of the enzyme in various kinds of hamster tissue. On immunohistological observation, glutathione peroxidase was localized mainly in the cytoplasm of the hepatooybes, epithelial cells of renal tubules, myooardial cells and skeletal muscle cells. Peripheral cells of the hepatic lobules showed stronger 'immunostaining' than those in the central zone.
