摘要
作者对新建立的一株小鼠T淋巴细胞性白血病克隆细胞株(LAC-1)生物学特性进行研究。结果表明:LAC-1细胞为Thy 1.1^+、Thy 1.2^-的分化早期T淋巴细胞性瘤细胞克隆,具有典型的淋巴细胞性白血病细胞形态学特征;它具有41超二倍染色体组型,电镜下可见胞浆内含小鼠白血病病毒颗粒,X-C检测为阳性;LAC-1细胞在体外培养中呈明显的白细胞介素2依赖性,体内致瘤作用严格受宿主的组织相容性限制,与母系L783白血病细胞相比,其致瘤活性明显减弱。LAO-1克隆细胞株的建立为我国T-S-Z小鼠白血病/淋巴瘤系统提供了一个新的体外研究模型。
A mouse T lymphocytic leukemio cell clone (LAC-1) was obtained from ascitic cells of L783 mouse leukemia by the limiting dilution method. The LAO-1 clone hag been continuously propagated in culture for 3 years, and gave the following biological characteristics: (1)Surface property and phenotype detection showed that LAO-1 cells were Thyl.1+, Thy1.2-, Lyt1-, Lyt2-, SmIg- and C3R- leukemio T cells. (2) Morphologically, typical lymphocytic leukemio cell characteristics were showed by light and electronmioroscope. (3) Proliferation response of LAC-1 cells showed an exponential growth rate when the cell culture was supplemented with IL-2. Using 3H-dT incorporation technique, the characteristic dose-dependence of IL-2 could also be demonstrated. (4) The cells multiplied 100 times within 6 days and the doubling time was 15.25 hrs. The peak of mitotio index was attained on day 2(58‰). (5) The number of chromosomes of LAO-1 cells were hyperdiploid with a modal number of 41. (6) MuL V particles were observed in the-cytoplasm of LAC-1 cells under the eleotronmicroscope. X-C assay and reverse transoriptase activity detection were both positive. (7)LAC-1 cells retained their carcinogenicity when they were inoculated intraperitoneally into syngeneio SW-1 mice and nude mice. Transplantation of LAO-1 cells to various other H-2 type mouse strains, however, resulted :in complete failure.
