摘要
小檗碱(Ber)1~4 mg·kg^1 iv明显提高兔的VFT,迷走神经切断或利血平3mg·kg^1 ip并不影响Ber提高VFT的作用,提示Ber抗心律失常的作用可能与自主神经无关,对兔在体心室肌细胞动作电位,Ber 1 mg·kg^1 iv的主要作用是降低V_(max),较高剂量(4 mg·kg^1)iv亦能延长APD_(90)和APD_(100),Ber1~4mg·kg^1 iv后,迷走神经或交感神经刺激引起的APD缩短作用减弱或消失,并抑制交感神经刺激诱发的DAD和触发活动,这些亦可能是Ber抗心律失常作用的机理。
Berberine(Ber)1-4 mg·kg-1 iv significantly and dose-dependently elevated the ventricular fibrillation threshold(VFT)induced by electric stimulation in rabbits.Reserpine and vagotomy exerted no influence on the increase in VFT with Ber,which implies that anti-arrhythmic action of Ber is most likely to bear upon its direct action on the heart without the involvement of the autonomic nervous system.The pronounced effect of Ber 1 mg·kg-1 iv on action potentials in rabbit myocardium in vivo was the reduction in the maximal velocity of depolarization(Vmax).At higher dose,Ber produced a prolongation in action potential duration at both 90% and 100% repolarization besides a more potent decrease in Vmax.After Ber 1 and 4 mg·kg-iv,vagal stimulation or left stellate ganglion stimulation could no longer cause the shortening of action potential duration at 30,50,90 and 100% repolarization,and the delayedafterdepolarization and triggered activity were remarkably inhibited and even abolished.Thus,the above-mentioned actions of Ber may contribute to its effectiveness against cardiac arrhythmias.
