摘要
本文通过建立具有亚临床病例特点的实验性甲亢和甲低模型,以及通过参照文献的方法建立应用荧光分光光度计测定血浆中普萘洛尔(Propranolol)的方法,研究了甲状腺功能状态对普萘洛尔药代动力学的影响,结果如下:对照组,甲亢组和甲低组大鼠ip普萘洛尔4mg/kg,求得其主要药代动力学参数——tl/2β(h);分别为4.54±0.65,3.58±0.35(P<0.05)和6.41±0.8(P<0.05);K_10(h^(-1)):分别为0.99±0.02,1.23±0.09(P<0.01)和0.66±0.08(P<0.001);AUC(ng/ml·h):分别为654.15±90.45,448.19±66.78(P<0.01)和1034.60±132.29(P<0.01);CL[mg/kg/h(ng/ml)]:分别为0.00622+0.00096,0.00911±0.00155(P<0.01)和0.00392±0.00058(P<0.01) Craax(ng/ml):对分别为501.46±40.13,408.33±57.65(P<0.01)和607.81±27 24(P<0.01)。
We use the experimental hyperthyroidism and hypothyroidsm model which possess sub - clinical characteristics, and we establismed the method to detrmine plasma contration of propranolol by use of fluorophotometer to study the effect of thyroid functional state on the pharmacokinetics of propranolol. The results are foliowing: Pharmacokinetic parameters of propranolol fowwowing ip 4mg/kg in contrlo-, hypertkyroidism and hypothyroidism group rats-t1/2β (h):4.54 ± 0.65,3.58 ± 0.35(P<0.05)and 6.41 ±0.8(P<0.05), respectuvely;k10(h-1):0. 99 ±0.02,1.23 ±0.09(P<0.01)and 0.66 ±0.08(P< 0.001) .respectively; AUC(ng/ ml+h): 654.14 ±90.45,448.19± 66.78(P<0.01)and 1034. 60 ± 132.29(P<0.01), respectively;CL [mg/kg/h/(ng/ml)]:0. 000622 ± 0. 00096, 0. 00911 ± 0. 00155(P< 0. 01)and 0. 00392 ± 0. 00058(P<0.01),respectively;Cmax(ng/ml):501.46 ±40.13, 408. 33 ± 57. 65(P<0.01 )and 607.81 ±27.24(P<0.01), respectively.
