石杉碱甲对D-半乳糖致衰老小鼠学习记忆行为的影响(英文)

摘要 背景:石杉碱甲是中国首创的一种可逆的胆碱酯酶抑制剂,其对良性记忆障碍有较好的疗效,能明显促进啮齿类动物的学习过程和记忆再现。目的:了解石杉碱甲对D-半乳糖拟衰老模型小鼠在跳台及迷宫实验中增强学习记忆行为的作用。设计:随机对照实验研究。单位:新乡医学院药物研究室。材料:实验在新乡医学院生理实验室及药物研究室完成。选择健康昆明种小鼠100只用跳台法实验和电迷宫法实验,每个实验组各50只。石杉碱甲片,由河南竹林众生制药股份有限公司提供,批号:031001;D-半乳糖,上海试剂二厂出品。干预:D-半乳糖对照组和石杉碱甲治疗组建立亚急性衰老模型:每日颈背部皮下注射120mg/(kg·d)D-半乳糖,连续6周,空白对照注射等量生理盐水。同时在以上处理的第37天开始,3个石杉碱甲治疗组分别用0.2mg/kg,0.4mg/kg,0.8mg/kg的3种不同剂量灌胃,空白对照组和D-半乳糖对照组以等量生理盐水灌胃,连续7d。主要观察指标:跳台法主要观察小鼠在跳台上安全区的潜伏时间和5min内小鼠触电次数;电迷宫实验主要观察小鼠逃至右侧电击区的错误次数。结果:跳台法检测结果显示,D-半乳糖致衰老模型小鼠的潜伏期时间较短,错误次数较多。空白对照组犤(9.2±38.7)s,(4.12±1.02)次犦与D-半乳糖对照组犤(49.7±27.1)s,(5.17±0.64) BACKGROUND:Huperzine A is a reversible choline enzyme(CHE) inhibitor initially developed in China,which has favorable therapeutic effectiveness in benign memory dysfunction,and could significantly promote the learning process and memory reappearance in rodent animals.OBJECTIVE:To comprehend the impacts of Huperzine A on the enhancement of learning, memory and behavior in platform and maze experiments in D Galoctose(Gal) induced senile mouse model.DESIGN:A randomized and controlled trial.SETTING:Institute of Pharmaceuticals of Xinxiang Medical College.MATERIALS:Study was performed in the Laboratory of Physiology and the Institute of Pharmaceuticals of Xinxiang Medical College from January to December in 2002.A total of 100 healthy Kunming mice were selected for platform and electric maze experiments with 50 mice in each experiment.Huperzine A tablet was obtained from Henan Zhulin Zhongsheng Pharmaceutical Company Limited(batch number:031001).D Gal is the product of Shanghai No.2 Reagent Factory.INTERVENTIONS:The establishment of subacute senile models in D Gal control group and Huperzine A therapy group:120 mg/(kg ·d) of D Gal was injected subcutaneously on the back of the mouse everyday for 6 weeks continuously.Same amount of saline was injected in blank control group.On the 37th day of the above management,mice of 3 Huperzine A therapy groups were treated with Huperzine A of three different dose:0.2 mg/kg,0.4 mg/kg and 0.8 mg/kg respectively through stomach perfusion,while same amount of saline was perfused in mice of both blank control group and D Gal control group for 7 days continuously.MAIN OUTCOME MEASURES:The latency in the safe area on the platform and the times of electric shock occurred within 5 minutes were observed in mice in platform experiment;the times of the mice mistakenly escaping to the right electric attacking area were observed in mice in electric maze experiment.RESULTS:①As revealed by platform experiment, the latency of D Gal induced senile mice was relatively short with more times of mistakes.There were significant differences between[(89.2±38.7) s,(4.12±1.02) times] of blank control group and [(49.7±27.1) s,(5.17±0.64) times] of D Gal control group(P< 0.05).The latency significantly prolonged, and the times of mistake significantly decreased in mice of Huperzine A therapy group (P< 0.01).②As revealed by electric maze experiment, the times of D Gal induced senile mice escaping to right electric attacking area significantly increased.Compared with[(7.02±1.06) times] of D Gal control group,the mistakes of blank control group [(6.05±0.70) times]was significantly less (P< 0.05),while the improvement of Huperzine A therapy group was even significant(P< 0.01) with the even less mistakes[(3.86±1.80) times] occurred in Huperzine A middle dose group(0.4 mg/kg).CONCLUSION:Huperzine A can effectively improve the learning and memory ability of D Gal induced senile mice, and there is dose effect relationship within certain range.
