摘要 在十八酸修饰的碳糊电极表面共价键合固定黄瓜ssDNA,制备了黄瓜DNA伏安传感器。在杂交液中,传感器表面的黄瓜ssDNA与杂交液中的ssDNA进行杂交反应时,电活性物质Co(bpy)3(ClO4)3配合物嵌入DNA双链中,使峰电流(△ip)增加。△ip与试液中DNA浓度成正比,可用于定量测定黄瓜DNA的含量。方法的线性范围为10~90ng/mL,检出限为2.97ng/mL。 The cucumber DNA voltammetric biosensor was prepared using the stearic acid-modified carbon paste electrode,on which the cucumber ssDNA was immobilized. In a hybridization solution,the cucumber ssDNA on the electrode hybridized with the ssDNA in the solution to form the dsDNA ,in which the complex Co(bpy)_(3)(ClO_(4))_(3) was intercalated. The complex yielded a voltammetric wave,which can be used to determine DNA. The linear range was from 0.010 to 0.090 μg/ mL with detection limit of 2.97 ng/mL.
