海南岛西瓜花叶病的病原鉴定 被引量:6

IDENTIFICATION OF PATHOGEN OF WATERMELON MOSAIC DISEASE ON HAINAN ISLAND

摘要 近年来,在海南岛发现了许多西瓜花叶病株,发病率一般为10~30%。通过人工接种5科18种植物,结果仅葫芦科9种植物表现花叶症状,藜科的苋色藜表现局部褪绿斑,普通烟、心叶烟等植物不表现症状。蚜虫可作为传毒媒介。病毒汁液的致死温度为55~58℃,稀释限点为10^(-3)~10^(-4),体外保毒期为1~4天。病毒粒子为柔性纤维状,长度为610~700毫微米。病叶超薄切片,在电子显微镜下观察到线状病毒粒子及感病细胞内风轮状和卷筒状内含体。鉴定结果表明,海南岛西瓜花叶病的病原属于西瓜花叶病毒2株系(WMV—2)。 Watermelon mosaic disease was found in watermelon, Citrullus vulgaris, on Hainan Island, with an incidence of 10—30%. Artificial transmission tests of the disease by sap inoculation on 18 species represanting 5 plant families showed that only 9 species in Cucurbitacese were infected. Yellow mottes on young leaves, Local lesions or local light—green mottings were induced in the leaves of Chenopodium amaranticolor and no symptoms appeared in Nicotabaeum and Nicotiana glutianosa etc The disease was readily transmitted by aphids The isolates from the diseased plants had thermal inactivation points 55—58℃, dilution end points 10^(-3)—10^(-4) and longevity 1—4 days. Electron microscope examination indicated that the isolated virus particles were flexuously filamentous with 610—700nm in length and oceured as cytoplasmic inclusion in pinwheel or roll shape. It is thus concluded that the pathogen of watermelon mosaic disease found on Hainan Island is Watermelon mosaic virus 2.
