摘要
本文报道坛紫菜Porphyra haitanensis、圆紫菜P.suborbiculata、铁钉紫菜P.ishigecota、海萝Gloiopeltis sp.的孢子在液氮超低温(-196℃)保存。试验表明二甲亚砜(DMSO)作为低温保护剂比甘油更好。冷冻方法有快速冷冻和慢速冷冻。用DMSO作为保护剂,快速冷冻时孢子存活率为30—40%,慢速冷冻时孢子存活率高达90—95%。在20—-60℃之间,样品进入液氮前慢速降温达到的温度越低,则孢子的存活率越高。经冷藏保存1天—5个月后,孢子存活率无显著变化,冷藏后的孢子萌发率正常,幼苗生长健壮。
The viability of spores of Porphyra haitancnsis, P. subcrbiculatc, P. ishigecota and one species of Gloiopeltis sp. after being frozen in liquid nitrogen was observed. It was indicated that dimethylsulfoxide was a better cryoprotectant for spores of red algae than glycerol.In the presence of DMSO (10-15%, V/V) most spores gave very high survival percentage(90-95%) when they were prefrozen slowly (0.5-1.0℃/min) to about - 60℃ before being immersed in liquid nitrogen and then rapidly thawed. However, the survival percentage was only 30-40% when rapidly frozen by direct immersion of spore materials in liquid nitrogen. The spores were slowly prefrozen to various temperatures at which they were plunged into liquid nitrogen. It was shown that the lower the temperature of the prefrozen spores, the higher the survival percentage was.The survival percentage of spores did not change significantly after they were stored for 1 day up to 5 months. After cryopreservation the spores were healthy and developed into normal sporelings.
