摘要
目的 研究梁金菇多糖 (LJPS)的抗肿瘤作用及机制。方法 采用四氮唑蓝比色法 (MTT法 )观察LJPS对K5 62细胞的增殖抑制 ,应用NBT试验和普通光镜观察细胞分化 ,流式细胞术 (FCM)和DNA凝胶电泳技术检测细胞凋亡。结果 LJPS对K5 62细胞有明显抑制增殖并诱导细胞凋亡、分化的作用 ,DNA凝胶电泳出现片断化的梯形带 ,FCM检测示细胞阻滞在G1期 ,出现凋亡峰。结论 LJPS对K5 62细胞具有抑制增殖、诱导凋亡和分化的作用。
Objective To investigate the anti-tumor effect and mechanism of Liangjin mushroom polysaccharide(LJPS).Methods Cell inhibitory was analyzed by MTT colorimetry cell differentiations were observed by morphology and nitroblue tetrazodium(NBT)test.Apoptosis was detected by flow cytometry(FCM)and DNA agarose gel electrophoresis.Results Liangin mushroom polysaccharide could significantly inhibit proliferation of K562 cell and induced differentiation and apoptosis of K562 cell exhibited a morphological characteristic of apoptosis and typical DNA ladder in DNA agarose gel electrophoresis,FCM analysis showed that cells stemed in the G 1 stage increased and the peak of sub-G 1 stage also increased in apoptosis.Conclusions Liangjin mushroom polysaccharide could significantly inhibit K562 cell proliferation,induce apoptosis and differentiations.
