摘要
用小鼠IL-3分泌细胞株WEHI-3和IL-3依赖细胞株32-D,获取了IL-3并对其活性进行了鉴定。在此基础上,选用国内实验性T-S-Z白血病系统中的一株小鼠淋巴瘤细胞克隆SAC-ⅡB_2,观察了IL-3对其增殖和分化的影响。结果表明,IL-3对SAC-ⅡB_2细胞有一短暂的刺激增殖作用;IL-3作用的SAC-ⅡB_2细胞未见成熟形态迹象出现,未见成熟T或B细胞表面标志Thy-1、Smlg的表达,也未显示成熟T、B细胞的功能。
Interleukin-3(IL-3) was initially defined as a factor in conditioned medium from Oon-A stimulated lymphocytes (Oon-A CM) that induces the enzyme 20-SDH in culture of nu/nu Splenic lymphocytes. IL-3 showed a number of activities, mainly the regulation of proliferation and differentiation of early hemato-poietio stem cells and a variety of lineage cells. An IL-3-containing supernatant from mouse myelomonocytic leukemia cell line WEHI-3 referred to as WEHI-3 conditioned medium (WEHI-OM) was used to determine the influence of IL-3 on development of mouse lymphoma cells SAO-IIB2. Experimental results indicated that DNA synthesis and 3H-TdE up-take by non-T and non-B SAO-IIB2 cells were increased significantly but transiently in the presence of IL-3. It was also shown that there were no morphological changes and no differentiated markers of mature T or B cells. All these suggested that IL-3 could enhance the proliferation of lymphoma cells and not the differentiation in mtro through out a short period of culture. The effects of IL-3 on differentiation of lymphoma cells were discussed.
