摘要
将纯化的猜传染性胃肠炎病毒免疫 BALB/C 小白鼠后,取免疫脾细胞与骨髓瘤细胞在 PEG 存在下进行融合,HAT-1640培养液筛选,并用 ELISA 间接法检测阳性克隆。通过有限稀释法进行三次克隆化后,获得了二株稳定分泌抗 TGEV 的单克隆抗体杂交瘤细胞株。杂交瘤细胞染色体皆在95条以上;其分泌抗体均属 IgG_1亚类。通过阻断试验证明其作用的特异性。对该单抗在诊断上的初步应用作了探讨。
Two stable hybridoma cell lines which secreted specific monoclonal antibodies have been obtained from the fussion of murine mycloma cells and spleen cells of BALB/C mice immu- nized with purified TGEV of swine.After selected by HAT-1640 culture sodium,the hybridoma cells have been cloned three times using limited dillution mcthod.The chromsome numbers of the two hybridoma cells were about 95,and the McAb of the two hybridoma cells wcre all of IgG_1 isotypcd by ELISA test.The Blocking test proved their specificity.The ap- plication of McAb in diagnosis was also sullessfully rcsearched.
