摘要
采用“酶—底物”荧光反应检测地面水中总大肠菌群的MPN值,是一种新方法。该方法主要由两部分组成:制备有利于总大肠杆菌群生长的LC培养基;选用能与总大肠菌群生成的β-半乳糖甙酶起反应的底物,4-甲基伞形酮-β-D-半乳糖甙。4-甲基伞形酮在3600A紫外线照射下具有强烈的蓝色荧光,根据荧光即可判别水样中是否含有总大肠杆菌群。此法与现行的标准“多管发酵法”进行平行实测比较,经t检验及相关性分析,结果基本一致,但在检测周期上,前者较后者可缩短2/3以上时间。
A new method for rapid enumeration of MPN value of coliform groups in surface water was described. This method was based on establishing a special me- dium as bacterial nutrient in combination with a spe- cial substrate for reaction with β--galactosidase releas- ed from coliforms. This rapid method was termed as 'LC mediun-Mllgal' system to compared with the me- thod of 'MM medium--ONPO' system extended recen- tly. The results indicated that the former was better than the later as shown by the ratio 'CFU units of coliforms colonies/CFU unite of heterotrophic bacteri- al colonies' screened from the culture with the two respective detective systems. The 'LC medium--Mllgal' system was also compared with the standard 'multi- ple--tube fermentation' method with large amount of surface water samples, no apparent difference was ob- served between these two methods on the basis of the test and correlation coefficient in Statistical analysis. On conclusion, the 'LC medium--Mllgal' system may be used to get faster result, it is sensitive. easier to ope- rate and required less labor and lower cost than the routine standard method.
