摘要
目的 利用早孕猕猴模型 ,了解米非司酮对腺垂体促性腺激素分泌的影响。方法 将 2 5只怀孕 38~ 4 5d的实验猕猴平均分为 5组 ,分别为不给予任何药物、给予安慰剂、米非司酮、米索前列醇及米非司酮配伍米索 ,取其腺垂体 ,应用单克隆抗体链菌素亲生物蛋白过氧化酶 (SP)免疫组织化学方法及电子计算机图像分析技术 ,观察促性腺激素分泌变化。结果 正常早孕猕猴促性腺激素位于腺垂体促性腺激素细胞胞质内 ,服米非司酮及服米非司酮配伍米索后 ,促性腺激素明显减少 (P <0 0 1) ;服用安慰剂及服米索前列醇无明显改变。结论 米非司酮作为口服抗早孕药物 ,可以直接作用于垂体 ,影响促性腺激素分泌。
Objective To examine the effects of mifepristone on gonadotropi n secretion at anterior pituitary by the model of early pregnant rhesus monkeys. Methods Anterior pituitary were obtained from pregnant rhesus m onkeys with gestation age of 38—45 days. Twenty-five monkeys were divided into 5 groups equally and were treated with nothing, placebo, mifepristone, misopros tol and mifepristone plus misoprostol respectively. The change of gonadotropin s ecretion was compared among 5 groups by using immunocytochemical histostainTM-S P method and computer image analysis technology. Results The cytoplasma of gonadotroph was stained obviously in normal early pregnant rhesus monkeys. Misoprostol and mifepristone plus misopros tol therapy significantly reduced gonadotropin content of gonadotroph (P<0 .01), while gonadotropin content had no significant change in placebo group and misoprostol group. Conclusion Mifepristone, as an anti-pregnant drug, might act d irectly at pituitary and affect gonadotropin secretion.
