自然衰老与半乳糖催老小鼠重要脏器的形态学差异 被引量:12

摘要 目的:观察自然衰老与半乳糖催老小鼠的病理形态学差异。方法:NIH2月龄小鼠27只,按体质量随机分为致衰老模型组,自然衰老组和对照组,每组9只。致衰老模型组每日皮下注射D-半乳糖100mg/kg,正常对照组每日皮下注射等量生理盐水,各持续42d。自然衰老组为常规饲养15月的小鼠。分别将上述各组小鼠断头处死后解剖,取心脏,肝脏,肾脏,肺脏,脑等脏器,常规病理取材,固定,脱水,包埋,苏木精伊红染色,制片,光镜下观察各主要脏器的病理形态学改变。结果:与正常对照组相比,自然衰老组和半乳糖催老组的各主要脏器的组织结构均有不同程度的老化改变;半乳糖催老组小鼠的肺组织中,肺泡腔面积未见明显减少,肺泡壁内部分毛细血管内皮细胞增生;自然衰老组小鼠的肺组织中,肺泡腔面积明显减少,部分实性变,肺间质纤维结缔组织增生,肺泡上皮细胞,毛细血管内皮细胞增生。半乳糖催老组小鼠肾脏的改变以肾小球数量减少,球系膜细胞增生为主;自然衰老组小鼠的肾脏以肾间质结缔组织增生和肾小管上皮细胞萎缩变性为主。心脏,肝脏,脑的病理组织学改变在自然衰老组和半乳糖催老组之间无明显差异。结论:肺脏和肾脏老化的病理组织学改变在自然衰老组和半乳糖催老组小鼠之间存在差异。 AIM:To observe the pathomorphological differences between natural senile mice and galactose induced aged mice.<METHODS:Totally 27 two month NIH mice were randomly divided into galactose induced aged group,naturally aged group,and control group according to their body mass.The mice in the galactose induced aged group were given subcutaneous injection with 100 mg/kg D galactose once a day;and the mice in the control group were given subcutaneous injection with the same amount of saline for 42 days respectively.While mice in the normal control group were fed with normal method, aged 15 months.All the mice of the groups were killed by decapitation.They were dissected, and the important organs like heart,liver,kidney,lung and brain were sampled.Materials were collected pathologically.Samples were fixed,dehydrated,embedded,haematoxylin eosin stained,produced,and histopathological changes were observed under light microscope.<RESULTS:Compared with the normal control group,structure of important organs in the naturally aged and galactose induced aged groups had different aging changes.As to lung tissue of mice in the galactose induced aged group,cavity area was not significantly decreased,capillary endothelial cells of alveolar wall interstitial portion was proliferated.On the other hand,for the mice in the naturally aged group,alveolar area was obviously decreased,portion was changed thoroughly,interstitial fiber connective tissues were proliferated,alveolar epithelium and capillary endothelial cells were proliferated.Kidney changes of mice in the D galactose induced group were featured as the decrease of glomerulus and the proliferation of mesengial;And those in the naturally aged group were featured as the proliferation of kidney interstitial connective tissues and the atrophy of renal tubule epithelial cells.No significant differences were found out in the pathologically histological tissue of heart,liver,and brain between D galactose induced aged group and naturally aged group.<CONCLUSION:Histopathologial differences of lung and kidney are found between naturally aged mouse and D galactose induced aged mouse.
