摘要
目的 观察左旋精氨酸 (L Arg)对大鼠自体移植静脉内膜增生的影响 ,探讨其作用机制。方法 建立大鼠自体静脉移植模型。分L Arg组和对照组。L Arg组于术前 7天开始用L Arg灌胃 (每天 1.0g kg)直至取材 ;对照组用等量生理盐水灌胃 ,于术后 1、2、4、6周及 8周取材。观测新生内膜与中膜面积比 (I M)、细胞间黏附分子 (ICAM 1)及细胞周期素A(CyclinA)的表达。结果 术后 2、4、6周及 8周I ML Arg组均显著低于对照组 (P <0 .0 1) ,术后不同时相ICAM 1及CyclinA表达水平较术前正常静脉均有明显增加 ,L Arg组ICAM 1及CyclinA蛋白阳性细胞百分比显著低于对照组 (P <0 .0 1)。结论 L Arg可以抑制移植静脉内膜增生病变 ,其机制与增加一氧化氮释放 ,减少单核细胞与内皮细胞的黏附 ,抑制血管平滑肌细胞增殖有关。
Objective To observe the effect of L-arginine on the intimal hyperplasia of autografted vein and to discuss mechanism.Methods Rat experimental models of autogenous vein graft were established.The rats were devided into L-arginine(L-Arg) group and control group.The L-Arg group was given L-Arg by gavage〔 1.0g/(kg·d)〕,which began 7 days before operation and continued until harvesting;Normal saline of the same volume was given to the control group.At 1,2,4 and 8 weeks after surgery,vein graft were harvested for the ratio of intimal area/media area(I/M)and expression of ICAM-1 and Cyclin A.Results At 2,4 and 8 weeks,the I/M value of L-Arg group was lower than that of the control group significantly(P<0.01).At different times after operation,there were obvious increasing extent of expression of ICAM-1 and Cyclin A,and the ICAM-1 and Cyclin A expression level in L-Arg group reduced as compared with control group(P<0.01).Conclusion L-Arg can inhibit intimal hyperplasia of graft vein.The protective effect may be related to the generation of nitric oxide to lessen monocyte adhesion to endothelial cell and inhibit smooth muscle cell proliferation.
