摘要
目的探讨β-连环蛋白(β-catenin)、细胞周期蛋白 D1(cyclinD1)在喉鳞癌中的表达及其生物学意义。方法采用免组化 SP 法检测58例喉鳞癌和10例正常喉上皮组织中的β-catenin、cyclinD1表达。结果 10例正常喉上皮β-catenin 表达位于细胞膜,胞浆未见表达,cyclinD1呈阴性表达。58例喉鳞癌中31例β-catenin 呈细胞浆表达,其中16例细胞核表达,27例细胞膜表达减少甚至缺失,而 cyclinD1细胞核内阳性表达率为58.62%。pN+的β-catenin 胞浆表达阳性率明显高于 pN_0(P<0.05),T_3~T_4喉癌病例中β-catenin 胞浆表达阳性率明显高于 T_1~T_2的喉癌病例(P<0.05),cyclin D1阳性表达仅与喉癌浸润程度有相关性(P<0.05),β-catenin胞浆表达、胞核表达与 cyclinD1胞核阳性表达存在一定正相关性。结论喉鳞癌中β-catenin 异常表达较为常见,表现在β-catenin 胞膜到胞浆、胞核的异位表达,且以细胞浆表达显著,β-catenin 异位表达与 cyclinD1阳性表达有一定正相关性。β-catenin 胞浆表达与喉鳞癌浸润程度、淋巴结转移有显著相关,检测喉鳞癌中β-catenin 表达可作为评估喉鳞癌恶性程度的重要生物学指标。
Purpose To explore the expression of β-catenin and cyclinD1 in laryngeal normal membrane(LNM)and laryngeal squamous cell carcinoma(LSCC).Methods By using immunohistochemical staining(SP-method),the expression of β-catenin and cyclinD1 were detected in 10 LNM and 58 LSCC.Results In 10 LNM,β-catenin showed high intense expression in the membrane and negative expression in the cytoplasm and nucleus.CydinDl showed negative expression in the nucleus.In 31 of 58 LSCC,β-catenin was expressed in the cytoplasm in carcinoma with varying degrees.27 of them accompanied by less,or even lost expression in the membrane in cancer samples.In 16 cases,β-catenin could be detected in the nucleus,but the positive expression of cyclinD1 was mainly confined to the nuclei(34/58).The positive rate of β-catenin expression in cytoplasm was significantly higher in those patients with pN+and T_(3-4)staging than the patients with pNo and T_(1-2) (P<0.05).but it had no correlation with the tumor differentiation and location(P>0.05).The positive expression of cyclinD1 had correlation with invasive depth,and the aberrant expression of β-catenin in cytoplasm and in nucleus was positively related to the expression of cyclinD1 in nucleus.Conclusion The aberrant expression of β-catenin occurred frequently in the LSCC,mainly including the translocation of β-catenin from membrane to the cytoplasm and nucleus,and the accumulation of β-catenin in cytoplasm was overt.The expression of β-catenin in cytoplasm and in nucleus was positively related to the expression of cyclinD1.And the aberrant expression of β-catenin was statistically con-elated to the lymph node and invasive depth.Detection expression of β-catenin could be used as an important biological index for evaluating the malignant degree of LSCC.
