摘要
对青枯菌(Pseudomonas solanacearum)产生颉颃作用的无致病力青枯菌菌株 nOE-104产生的细菌素效价为128AU/ml;丝裂霉素 C 可将其效价提高至512AU/ml;UV 照射无诱导效应.CPG 培养液中产生的 nOE-104细菌素粗提液,经硫酸铵沉淀,沉淀物于磷酸钠缓冲液中透析后,通过 DE-52离子交换柱分离纯化.纯化的 nOE-104细菌素具有生物活性,分子量为19100;电镜下观察不到其粒体;对热不稳定,50℃处理10min 即失活;有机溶剂如甲醇、氯仿等对其生物活性无影响;最适pH 为6~8.蛋白酶 K 可以完全破坏 nOE-104细菌素活性,而核酶、胰蛋白酶和溶菌酶对其活性无影响.该细菌素具有蛋白质性质,含有16种氨基酸.
An avirulent,bacteriocin-producing strain nOE-104 of P.solanacearum could inhibit viru-lent strain of P.solanacearum.The titre of bacteriocin from strain nOE-104 is 128AU/ml.Mitomycin Ccould raise the bactrrioein titre more than four times.But UV could not in(?)rease the bacterioein titre.Thecrude bacteriocin could be purified by chromatography on DE-52.It has a molecular weight of 19 100.Theparticle of bacterioein from strain nOE-104 is invisible under electron mieroscope.This bacteriocin is sensi-tive to heat above 50℃,but insensitive to some organic-compounds,such as methanol,ethanol,and chloro-form.The optimum pH for the activity is between 6~8.The activity of bacteriocin from strain nOE-104 is complctely destroyed by Pronase K,but not byTrypsin,DNase,RNase,Lysozyme.This bacteriocin is a protein consist ing of 16 kinds of amino acids.
