摘要
应用间接免疫荧光技术,发现肝片形吸虫头部咽的两侧各有一强的5-羟色胺(5-HT)荧光集落(脑神经节),由其发出的神经干分布于虫体的背腹及两侧,神经干继续发出细小分支直至皮肌层和吸盘等.生殖系统(睾丸、卵巢、子宫、卵模和卵黄腺纵隔等)和消化系统(咽和肠管壁等)亦有5-HT 免疫反应分布.揭示肝片形吸虫体内不仅存在5-HT,而且存在色胺能神经系统,证明了5-HT 为肝片形吸虫的神经递质.
In rabbits antisera were raised against an antigen prepared by coupling 5-HT to bovine serumalbumin (BSA) using formaldehyde as a coupling reagent.Fresh adult Faseiola hepatica were fixed with 4%formaldehyde and sectioned on a eryostat (-25℃).The sections were stained by indirect immunofluoreseencetechnique.Abundant immunofluorescen(?)e specific for 5-HT was observed in bilateral ganglia proximal topharynx.It was demonstrated from the specific immunofluorescenee that symmetrical serotonergie nervetrunks bilaterally run from the aforemetioned ganglia and joined numerous lateral branches which extend finenerves innervating the tegumental syncytium.The yellow fluorescence of 5-HT was also found in thetegument,the parts surrounded pharynx and sucker,testes,ovary,uterus,vitellaria and its longitudinalseptum,ootype and gut walls,etc.The distribution of 5-HT within the nervous system strongly implied that5-HT was functioning as a neurotransmitter in Fasciola hepatica.
