苎麻疫霉卵孢子萌发研究 被引量:5

摘要 苎麻疫霉(Phytophthora boehmeriae Sawada.)菌株在利马豆培养基平板上形成的卵孢子,用KMnO_4和 H2O_2处理,在 S+L 培养基上诱导萌发(25℃).结果表明,用 KMnO_4和 H_2O_2处理卵孢子均能刺激其萌发.在一定时期内卵孢子萌发率随卵孢子保存时间的增加而上升,保存35d 的卵孢子用0.3%H_2O_2处理2min 和保存45d 的卵孢子用0.1%KMnO_4处理20min,萌发率最高,分别为80%和92%.卵孢子萌发时给予光照对萌发有明显刺激作用,卵孢子在萌发过程中的光照条件以黑光灯连续照射效果最好,日光灯连续照射效果较差.所测定的4种培养基中以 S+L 对卵孢子的萌发效果最好,其次为SA 和 WA. Oospores of Phytophthora boehmeriae were treated with KMnO_4 and H_2O_2 respectively,andgerminated on S+L medium for 12 days.Results showed that the germination of oospores was stimulated bythe pretreatment of oospores with those two oxidizers.The treatment with KMnO_4 and H_2O_2 also preventedthe germination of sporangia and recidual mycelial fragments.Germination rate increased with the age ofoospores,reaching about 80% at 35-day-old and 92% at 45-day-old when oospores treated with 0.3%H_2O_2 for 2min,and with 0.1% KMnO_4 for 20min,respectively.Illumination was essential for the germi-nation of oospores on S+L medium.The germination rate reached more than 80% when oospores were ex-posed to light and only 0~6% when light omitted.The best illumination for oospore germination was blacklight(near UV.),and the best medium was S+L.
