摘要 目的分析骨髓基质干细胞向神经元样细胞分化前后相关基因的表达。方法用β-巯基乙醇诱导骨髓基质干细胞向神经元样细胞分化,诱导后8h,提取诱导前后的骨髓基质干细胞mRNA,进行微管相关蛋白-2(MAP-2)、生长相关蛋白43(GAP-43)、神经元特异性烯醇化酶(NSE)、中间丝nestin和神经丝(NF)基因的RT-PCR,观察其诱导前后的表达。结果NSE在诱导前后均有表达,MAP-2、GAP-43、nestin和NF诱导前无表达,诱导后有表达。结论骨髓基质干细胞向神经元样细胞的分化与MAP-2、GAP-43、nestin和NF可能有关。 AIM: To analyze neuron-related gene expression before and after mesenchymal stem cells (MSCs) differentiating into neuron-like cells. METHODS: MSCs were induced to neuron-like cells with β-mercaptoethanol. Before inducement and at 8 h after inducement, the total RNA was extracted, then the expression of microtubule-associated protein-2 (MAP-2), growth-associated protein-43 (GAP-43), NSE, nestin and neurofilament (NF) mRNA were detected with RT-PCR. RESULTS: NSE mRNA expressed before and after inducement, MAP-2, GAP-43, nestin and NF mRNA only expressed after inducement. CONCLUSION: The differentiation of MSCs into neuron-like cells may be related to MAP-2, GAP-43, nestin and NF expression. [
