HCA587基因在肝细胞癌中的表达及其生物信息学分析 被引量:1

摘要 目的 :本研究检测肝细胞癌抗原 5 87( HCA5 87)基因 m RNA在肝细胞癌 ( HCC)中的表达情况 ,结合病人临床资料进行分析 ,探讨 HCA5 87基因与 HCC病人临床指标及转移与复发的关系 ,为 HCA5 87基因编码蛋白用于 HCC治疗提供依据。方法 :用反转录 -聚合酶链反应 ( RT-PCR)方法对 HCC病人的癌组织和相应癌旁组织及对照 (肝硬化和正常肝组织 )中 HCA5 87m RNA的表达进行了检测 ,为验证结果的可靠性 ,随机选取RT-PCR呈阳性的产物 4例进行 DNA序列测定 ,结合 AFP、抗 HCV、HBs Ag、肿瘤直径、分化程度等临床指标进行分析。并通过互联网对 HCA5 87进行生物信息学分析。结果 :46例 HCC病人中 ,HCA5 87的阳性表达率为3 2 .6% ( 1 5 /4 6) ,相应的癌旁组织中均为阴性 ;所测的 1 0例肝硬化和 1 0例正常肝组织均未检测到 HCA5 87的表达。 DNA测序结果表明 RT-PCR产物确为 HCA 5 87c DNA。 HCA5 87的表达与年龄、性别、肿瘤大小、分化程度、血清甲胎球蛋白 ( AFP)水平、HBV和 HCV感染无显著相关性 ( P >0 .0 5 )。在部分 AFP正常 ( <2 5 ng/L)HCC病人中存在 HCA5 87基因的表达。HCA5 87与 MAGE C2、MAGE E1、CT1 0的分子有很大的同源性 ;它精确定位于染色体 Xq2 7.2上 ,HCA5 87是一种新的 MAGE家族中的 CT抗原。结论 Objective: To investigate the expression of four hapatocellular cancer antigen587 (HCA587) gene mRNA in hepatocellular carcinoma and to explore the possibility of HCA587s gene encoding protein used as a target for immuno-therapy in HCC patients. Methods: The expression of HCA587 mRNA was detected using RT-PCR in HCC tissues and the corresponding adjacent non-HCC tissues from 46 HCC patients, 10 samples of cirrhosis and 10 samples of normal liver tissues. 4 samples selected randomly from HCA587 PCR positive products were sequenced. The relationship between positive expression rate of HCA587 gene and clinical and lab data including AFP, anti-HCV, HBsAg and the diameter of the tumors in HCC patients was also determined. Bioinformations of HCA587 were analysed at World Wide Web. Results: Of 46 HCC tissues, HCA587 mRNA was detectable in 37.2%(15/46)of HCC samples. No expression of HCA587 could be detectable in the corresponding adjacent non-HCC tissues. In addition, none of 10 samples of cirrhosis and normal liver tissue expressed each HCA587 gene mRNA. The DNA sequence confirmed that the RT-PCR products were truly HCA587 cDNA. No relationship was found between the expression of HCA587 and clinical indicators such as age, sex, tumor size, degree of tumor differentiation, serum α-fetoprotein level and infection of hepatitis B virus or hepatitis C virus($P$>0.05). However, in some patients with normal serum α-fetoprotein (<25 ng/L), specific expression HCA587 genes was observed. The genomic structure of HCA587 and MAGE C2、MAGE E1、CT10 are highly homologous to each other. The chromosomal localization of HCA587 gene is in Xq27.2. These results suggest that HCA587 may be a new cancer-testis antigen and belong to melanoma-associated antigen (MAGE) family. Conclusion: HCA587 gene mRNA is expressed with a high percentage and specificity in hepatocellular carcinomas and their products are new potential promising targets for antigen-specific immuno-therapy of HCC. No relationship was found between the expression of HCA587 and clinical indicators such as age, sex, tumor size, serum α-fetoprotein level and infection of hepatitis virus. In some cases with normal serum α-fetoprotein, specific expression of HCA587 genes was observed in HCC, suggesting applying its mRNA as tumor markers to detect circulating HCC cells as adjuvant diagnostic tool and as indicators of reccurrence and prognosis.;
