摘要
通过大鼠缺锌(ZD)、过量锌(ZE)模型,研究锌对脾脏T、B淋巴细胞功能的影响。结果:①ZD、ZE组的脾重及脾脏指数下降,与对喂(PF)组、自由喂养(AL)组、缺锌后再补锌(ZD+AL)组比较,差别非常显著。②ZD、ZE组外周血白细胞减低,以淋巴细胞为主。③ZD、2E组T、B淋巴细胞增殖率非常显著低于PF、AL和ZD+AL组。④ZD、ZE组的总脾淋巴细胞,T、B淋巴细胞细胞周期G0/G1%增高,而S%、G2+M%和增殖指数下降,与PF、AL和ZD+AL组比较,差别非常显著。⑤ZD、ZE组脾脏T淋巴细胞亚群CD3、CD4、CD8和CD4/CD8比值均非常显著地低于PF、AL和ZD+AL组,尤以CD4降低显著。
The effects of zinc on lymphocytic function of spleen on zinc-deficient (ZD) and zinc-excessive (ZE) feed were observed. The findings were as follows;
1. The weight of spleen and spleen fragility index were significantly lower in ZD and ZE rats than in those of paired feeding(PF) group or adequate libitum(AL) group. They restored the normal level after zinc repletion for the ZD animals(ZD+AL) groups.
2. The leucocytic count of the peripheral blood .especially the proportion of the lymphocytes of ZD and ZE rats were significantly lower than that of PF.AL and ZD+AL groups.
3. The proliferative responses of the splenic lymphocytes to ConA and to LPS of ZD and ZE rats were significantly much lower than those of PF. AL and ZD+AL groups.
4. Analysis of cell cycle of splenic lymphocytes was performed with flow cytometry. It was found that GO/G1 phase of total lymphocyte.T and B lymphocyte was significantly higher and S.G2+M phase lower in ZD and ZE rats than in those of PF.AL and ZD+AL.
5. Analysis of T lymphocytic subgroups were performed with flow cytometry. It was found that CDS, CD4.CD8.CD4/CD8 were significantly lower than in those of PF.AL and ZD+AL,especially in CD4.
