NSE-McAb化学发光酶免疫分析法的建立及应用

Establishment of Two-site Sandwich Chemiluminescence Enzyme Im- munoassay of NSE-McAb and Applied in the Small Cell Lung Cancer

摘要 本文在自制8株神经元特异性烯醇化酶单抗(NSE-McAb)的基础上,对4株McAb分别标记辣根过氧化物酶(HRP),经多次方阵排列滴定,建立了NSE-McAb双位点夹心化学发光酶免疫分析法(CLEIA)。通过对23例SCLC,32例NSCLC,22例BPD和正常对照血清中NSE检测表明,对SCLC诊断的阳性率为78.3%,特异性为93.0%,对SCLC的临床分期和判断预后也有一定的价值。 We established two-site sandwich chemiiuminescence enzyme immunoassay (CLEIA) and preformed multiple checkboard titration, using HRP separatelly labeling 4 McAbs which purifice 8 McAbs by our laboratory. We measured the serum NSE levels of 23 patients with SCLC. 32 patients with non-small cell lung cancer (NSCLC).22 patients with benin pulmonary disease(BPD) and normal controls(NC). The positive rate of detectable was 78. 3% and specific was 93. 0% with SCLC. Two-site sandwich CLEIA may be valuable for clinical staging and evaluating prognosis in patients with SCLC.
