PHA对人扁桃体T细胞CD4和CD8抗原复合表达的研究 被引量:5

摘要 本文利用双色荧光试剂、流式细胞术证明了新鲜分离的儿童扁桃体T细胞中含有少量的CD4^+CD8^+细胞。CD4^+CD8^+的复合表达似乎与T细胞活化有关,其主要表达在大的淋巴母细胞上。用PHA刺激T细胞5天后,产生的CD4^+CD8^+细胞数可达38.33%;与此同时,细胞上CD8抗原的密度明显增高。另外,PHA活化的CD4^+CD8^+细胞能高水平表达CD25、CD38及HLA-DR抗原,但缺乏CD1抗原的表达。我们的实验结果提示:人扁桃体T细胞活化产生的CD4^+CD8^+细胞亚群不同于CD4^+CD8^+的所谓双阳性胸腺细胞,由于CD4和CD8分子直接与MHC抗原相互作用,这些分子的复合表达可能在免疫调节中起重要作用。 Using two-color fluorescence flow cytometry, we were able to detect the presence of small numbers of CD4+CD8'l'cells(3. 29±1. 14%)in freshly isolated T cell populations derived from human tonsil. Coexpression of CD4 and CDS was predominantly found on large blastlike cells and appeared to be related to activation. Stimulation of T cells with PHA for 5 days resulted in the significant increase of CD4','CD8+ cells. Coexpression was accompanied by significant increase of CDS antigenic densities. The PHA-activated CD4+ CDS'1' cells expressed high levels of activation antigens CD25.CD38 and HLA-DR,but lacked GDI antigen. Our data suggested that tonsillar T cell activation results in the generation of CD4+CD8+ cells which is distinct from thymic cells. Because CD4 and CDS molecules may interact directly with MHC antigens. Coexpression of these molecules may have an important role in immune regulation.
