摘要
目的 :探讨肿瘤坏死因子 (TNF -α)与干扰素 (IFN -γ)联合应用对体外乳鼠雪旺氏细胞增殖的抑制作用。方法 :建立体外乳鼠雪旺氏细胞培养体系 ;经分离、纯化、传代的雪旺氏细胞通过免疫组化、电镜鉴定后 ,纯度达到90 %以上后进行实验。实验分为空白组、对照组、TNF -α组、IFN -γ组及TNF -α +IFN -γ组 ,分别作用 72h后 ,应用四氮唑蓝 (MTT)比色法测定各组的OD值 ,计算其抑制率。结果 :TNF -α和IFN -γ对乳鼠雪旺氏细胞生长增殖均有直接抑制作用 ,其中TNF -α在 10 0~ 2 0 0 0U/ml浓度范围内对正常雪旺氏细胞增殖有抑制作用 ,呈剂量依赖关系 ,在10 0 0U/ml抑制率达到最大。联合用药能明显提高TNF -α对乳鼠雪旺氏细胞生长增殖的抑制效应。结论 :TNF -α对乳鼠雪旺氏细胞生长增殖起抑制作用 ,并呈剂量依赖性。IFN -γ与TNF -α联合用药有明显的协同抑制效应。
Object To investigate the synergisti c effect of tumor necrosis factor-α(TNF-α) and interferon-γ(IFN-γ) on the prol iferation of newborn rat Schwann cells in virto. Methods N ewborn rat Schwann cells were cultured in virto. Then the cultured and purified Schwann cells were identified with immunohistochemistry and electron microscopy. The percentage of purified Schwann cells was more than 90%. Schwann cells were cultured in the medium containing different concentrations of TNF-α、IFN-γ or TNF-α and IFN-γ from newborn Wistar rats and incubated for 72h, respectively. MTT colorimetric assay was used to study the effects of TNF-α,IFN-γ or TNF-α and IFN-γ on Schwann cell growth. Results The proliferati ve responses of Schwann cells were directly inhibited by TNF-α or IFN-γ. TNF- α inhibited proliferation in dose dependent manner in the levels from 50 U/ml to 2 000 U/ml. The inhibitory effect can reach maximum when the concentration in 1 000 U/ml. However, A synergistic greatly increase in the inhibitory effects o f TNF-α and IFN-γ. Conclusion TNF-α inhibited Schwann cells proliferation in dose dependent manner. There was a synergistic increase i n the inhibitory effects of TNF-α and IFN-γ.
