基质金属蛋白酶-2及基质金属蛋白酶组织抑制剂-2在卵巢恶性肿瘤中的表达及其意义

摘要 目的 :探讨基质金属蛋白酶 - 2 (MMP- 2 )及基质金属蛋白酶组织抑制剂 - 2 (TIMP- 2 )在卵巢肿瘤中的表达及与卵巢恶性肿瘤临床病理特征和预后的关系。方法 :应用 RT- PCR方法检测 4 8例卵巢恶性肿瘤、2 1例良性卵巢肿瘤及 2 2例正常卵巢组织中 MMP- 2 m RNA及 TIMP- 2 m RNA的表达情况 ,进行阳性率及半定量的比较 ,并将其结果与临床及病理学资料进行统计学分析。结果 :MMP- 2在卵巢恶性肿瘤组织中的表达水平 (0 .92 8± 0 .796 )明显高于正常卵巢组织 (0 .5 2 7± 0 .5 78) (P <0 .0 5 )。 TIMP- 2在卵巢恶性肿瘤及良性肿瘤中阳性率及半定量 (分别为 75 .0 %、0 .777± 0 .6 5 3、76 .2 %、0 .771± 0 .6 35 ) ,明显高于正常卵巢 (分别为 4 5 .5 %、0 .382± 0 .6 33) (P <0 .0 5 )。其表达水平与卵巢恶性肿瘤的临床病理特征无相关性。多因素生存回归分析显示 MMP- 2、TIMP- 2可作为卵巢恶性肿瘤的独立预后因素。结论 :MMP- 2及 TIMP- 2可能与卵巢恶性肿瘤的发生发展有关。 Objective:To investigate the relationship between the expressions of matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of matrix metalloproteinase-2(TIMP-2) mRNA and clinicopathologic characteristics of patients with ovarian malignant tumor.Methods:RT-PCR was used to detected the expression of MMP-2 and TIMP-2 mRNA in 48 cases of ovarian malignant tumor tissues,21 cases of benign ovarian tumor tissues and 22 cases of normal ovarian tissues .Result:The half-quantity of MMP-2 mRNA was significantly higher in ovarian malignant tumor than that in normal ovarian tissue(P<0.05).The relative value and positive rates of TIMP-2 is significantly higher in ovarian malignant tumor and benign tumor than those in normal ovarian tissue .The expression of MMP-2 and TIMP-2 is not significantly relevant to clinicopathologic characteristics of patient with ovarian malignant tumor. Multivariate regression survival analysis show MMP-2, TIMP-2 may be independent prognosis factor.Conclusion:The overexpression of MMP-2 and TIMP-2 may contribute to the development of ovarian malignant tumors.
