摘要
目的 观察保肝中药复方药物血清对传代肝星形细胞组织金属蛋白酶抑制剂 (TIMP 1)表达及凋亡的影响 ,以探讨其抗肝纤维化的作用靶点。方法 采用大鼠肝星形细胞分离培养 ,免疫组化ABC法检测肝星形细胞TIMP 1表达与药物血清的干预 ,经图像分析定量 ;Giemsa染色和TUNEL染色计算凋亡细胞百分比。结果 药物血清组的肝星形细胞TIMP 1阳性反应物吸光度与对照组比P <0 0 5 ;细胞凋亡率为 10 3%。结论 保肝中药复方药物血清能抑制肝星形细胞TIMP 1的表达 ,并促进活化的肝星形细胞凋亡。
Objective To observe the effects of liver-protective TCM drugs on the expression of TIMP-1 in HSC and HSC apoptosis on the purpose of investigating their target points for resisting hepatic fibrosis.Method First, the rat HSC was isolated and cultivated, and then, TIMP-1 expression in HSC and the interfering effects of the TCM drug-medicated serum on the TIMP-1 expression were detected by the ABC immunohistochemical method, the results of which were analyzed by a quantitative image analytic technique; and the percentage of HSC apoptosis was calculated by the Giemsa staining method and TUNEL staining method. Results There was a significant difference between the absorbency of TIMP-1 positive reactants in hepatic HSC in the TCM drug-medicated serum group and that in the control group (P<0.05); and the apoptotic rate was 10.3% in the TCM drug-medicated serum group. Conclusion The serum mediated with TCM liver-protective drugs could inhibit the TIMP-1 expression of hepatic HSC and promote apoptosis of the activated hepatic HSC.
