内毒素血症大鼠心肌炎性介质产生及组织学变化 被引量:1

摘要 目的 :研究内毒素血症 (ETM )大鼠血循环及心肌内重要炎性介质 (TNF α和IL 1β)的产生和变化情况 ,以及心肌组织形态学变化。方法 :随机将大鼠分为正常对照组、ETM组 (由尾静脉按 3mg/kg注射LPS引发ETM )。采用放射免疫分析法检测不同时间点血循环及心肌内TNF α和IL 1β含量。再通过病理形态学技术 ,观察心肌组织形态学改变。结果 :正常大鼠血循环及心肌内含有一定量TNF α和IL 1β。与正常对照组相比 ,ETM发生后 ,血循环及心肌内TNF α和IL 1β含量均有不同程度的升高 (P <0 0 5或P <0 0 1)。病理形态学观察表明 ,ETM发生后 ,大鼠心肌组织表现出炎症改变 ,主要为充血、出血、渗出及炎性细胞浸润 ,以伤后 2 4h更为明显。结论 :内毒素可致血循环及心肌内TNF α和IL 1β过度分泌 ,并使心肌出现炎症反应。即内毒素诱导的TNF α和IL 1β介质对于ETM心肌发病起重要作用。 Objective To investigate the secretion and changes in rats with TNF α and IL 1β, and pathomorphological change in myocardium. Methods Rats were randomly divided into two groups: normal control and Endotoxemia (ETM) (ETM rats mere made by tail venous injection of LPS at the 3 mg/kg). The radioimmunoassay method was used to detect the level of TNF α and IL 1β in the blood circulation and myocardium of ETM affected rats. The inflammatory change in myocardium was observed by pathomorphological method. Results Normal rats had a definite amount of TNF α and IL 1β in the blood circulation and myocardium. The level of TNF α and IL 1β rose significantly in ETM rats ( P <0 05~0 01). Also the myocardium of ETM rats has shown inflammatory changes, mainly in the form of hyperaemia, bleeding, exudation, and the infiltration of inflammatory cells, which were more serous at 24h after the injury. Conclusion The endotoxin can lead to the excessive secretion of the TNF α and IL 1β in the blood and myocardium of ETM affected rats, namely, both TNF α and IL 1β are highly related to the ETM incidence and very likely to be the important inflammation mediators in the pathological process of ETM.
