体外诱导成人骨髓基质细胞分化为神经元样细胞

摘要 目的探讨成人骨髓基质细胞(BMSCs)的生物学特性及诱导为神经元样细胞的方法。方法采用Ficoll-Paque液(1.077×103g/L)从成人骨髓中分离BMSC,体外扩增,以碱性成纤维细胞生长因子(bFGF)、表皮生长因子(EGF)、全反式维甲酸(ATRA)、联丁酰基环腺苷酸(dbcAMP)为诱导剂,诱导期间观察细胞形态和数目的变化,并通过免疫细胞化学鉴定神经元烯醇化酶(NSE)、巢蛋白(nestin)、胶质纤维酸性蛋白(GFAP)的表达情况。结果此诱导条件下95%的细胞成为神经元样细胞,免疫组织化学检查显示这些细胞表达NSE、GFAP或nestin。结论bFGF和EGF、ATRA、dbcAMP能诱导成人BMSCs分化为神经元样细胞,而且具有较高的阳性转化率。 Objective To explore the biological characteristics of adult bone marrow stromal cells (BMSCs) and the methods of inducing differentiation them into neuron-like cells in vitro. Methods BMSCs were isolated from adult bone marrow with density gradient with Ficoll-paque (1.077×103g/L) and expanded in vitro. BMSCs were treated by basic fibroblast growth factor (bFGF), epidermal growth factor (EGF), trans retinoic acid (ATRA), and dibutyryl cylicAMP (dbcAMP). Immunohistochemistry was used to identify the cell expression of neuron specific enolase (NSE), glial fibrillary acidic protein (GFAP) and nestin. Result Under this condition, 95% of the cells differentiated into neuron-like cells. The cells treated with these agents expressed NSE or nestin or GFAP. Conclusion bFGF?EGF?ATRA?dbcAMP can induce human BMSCs to differentiate into neuron-like cells in vitro, and the percentage of the neuron-like cells is rather higher.
