摘要
目的 :观察川芎嗪 (TMP)对顺铂 (DDP)所致大鼠肾毒性的保护作用。方法 :DDP单次 8mg/kgip诱导大鼠肾损伤 ,于给DDP 2天前开始分别ipTMP 5 0mg/kg、10 0mg/kg ,连续 5天 ,末次给药 4小时后处死大鼠 ,计算肾脏指数、测血清尿素氮 (BUN )和肌酐 (Scr)含量 ,测 2 4小时尿蛋白含量及尿N 乙酰 β D 氨基葡萄糖苷酶 (NAG)、γ 谷氨酸转换酶 (GGT)、碱性磷酸酶 (ALP)的活性 ;光镜观察肾组织形态学变化。结果 :TMP呈剂量依赖性降低DDP所致大鼠肾脏指数 ,BNU、Scr含量 ,2 4小时尿蛋白含量及尿NAG、GGT、ALP活性的升高 ;肾脏病理组织学检查可明显改善DDP引起的肾小球充血、肾小管浊肿及管型的病理改变。结论
Objective: To investigate the preventive effect of ligustrazine on cisplatin induced nephrotoxicity in rats. Methods: Nephrotoxicity in rats was induced to be impaired with single ip cisplatin at dose of 8mg/kg. Ligustrazine (50mg/kg·d、100mg/kg·d) was given two days before the cisplatin injection for five days. Then the rats were sacrificed four hours after the last injection. After that Kidneys were removed to calculate the renal index, Blood urea nitrogen(BUN) and serum creatinine (Scr) was tested , urine samples obtained to find the content of 24 hours urinary protein and the activity of N acetyl β D glucosaminidase(NAG) γ Glutamyl transpeptidase(GGT) and alkaline phosphatase(ALP). Optical microscope was employed to observe the structure of renal. Results: the ligustrazine administration significantly renders a decline of the rise of the renal index (p<0.05),the content of BUN Scr(p<0.05,p<0.01),the content of 24h urinary protein and activity of urinary NAG GGT ALP induced by cisplatin, and it is dose effect relationship. The histopathological changes of kidney suggest that the ligustrazine can notably improve the congestion of glomeruli, swelling of kidney tubules and the casts in tubular lumen caused by cisplatin. Conclusion: Ligustrazine can prevent against the nephrotoxicity induced by cisplatin.
