摘要
将碱性磷酸酶抗碱性磷酸酶桥联酶标方法(APAAP)的着色细胞以积分表示,建立了APAAP法的半定量加权积分法。本法测定的单抗MGd1在KATO-Ⅲ细胞上表达的线性关系10^(-8)～5×10~2mol/L呈现良好的线性关系(r=0.9836)。本法定量可靠,简便易行,灵敏度高。应用该法对人胃癌细胞单克隆抗体MGd1和MGb2及其与抗癌药喜树碱偶联物的活性进行了评定,取得了良好的结果。
The qualitative cytochemical reaction of alkaline phosphatase anti-alkaline
phosphatase (APAAP) was modified by changing the score evaluation into a relatively quantita-
tive method. The immunoreactivity of MGd_1(monoantibody of antigen from KATO-Ⅲ cell) with
KATO-Ⅲ cell line was determined by this method. The relationship between MGd_1 concentra-
tion and immuno-reactivity was linear (r=0.9836). The advantages of this method are quantita-
tive reliable, rapid, simple and sensitive.
