摘要
全胚胎体外培养技术是近年来发展起来的一项难度较大的新的实验方法,在胚胎学、畸胎学和毒理学等研究领域具有重要的应用价值.本文详细讨论了此项技术的建立及评价.
The technique of whole embryo culture was that 9.5-day rat embryos are transplanted from the uterus to culture bottle and cultured in vitro.The medium was serum centrifugalized immediately after withdrawal from the donor rat,heat inactivated in 56℃ waterbath for 30 minutes and gassed.The culture bottle was filled with 5% CO_2 for first 48 hours,then with 5% CO_2,40% O_2 for another 24 hours.During culture,the culture bottle was rotated continuously 15～20 times per minute.At the 72th hour we observed the development of embryos through the yolk sac,circulation,rotation,heartbeat,neural tube,visceral arches,fore and hind limbs,somites and total protein of embryo.Through comparison of the development of the embryo in vivo and in vitro,we found that during the first 24 hours of the culture the rate of the development of embryos was similar in both conditions,but after 48 hours of the culture the development of embryo in vitro was slower than in vivo.At the 72th hour of the culture,the differentiation and formation of main oragns and systems of embryos were completed in both conditions.The whole embryo culture is a good method in research of embryology,teratology,toxicology.
