摘要
目的 观察糖基化终产物 (AGEs)对人肾系膜细胞 (HRMC)分泌白介素 8(IL 8)的影响。方法 常规制备糖基化终产物 (AGEs BSA) ,干预体外培养的HRMC ,用ELISA法检测培养上清中IL 8浓度。结果 不同浓度AGEs BSA(16 0、32 0和 6 4 0 μg/ml)干预HRMC 4 8h ,上清中IL 8水平分别为 178 1± 15 6、4 75 4± 18 9和 2 4 85 2± 99 9pg/ml(P <0 0 1) ;AGEs BSA(6 4 0 μg/ml)干预HRMC4h后IL 8分泌量开始升高 (71 9± 9 2 pg/ml) ,并随时间延长 (8、2 4和 4 8h)进一步升高 ,分别为 5 4 6 6± 4 9 7、6 5 2 1± 4 2 8和 2 4 85 2± 99 9pg/ml(P <0 0 1)。 结论 AGEs BSA呈浓度和时间依赖方式增加HRMC分泌IL
Objective To study the effect of advanced glycosylation end products(AGEs)on the expression of interleukin-8(IL-8)in cultured human renal mesangial cells(HRMC).Methods The AGEs-BSA was prepared by incubating bovine serum albumin(BSA)with glucose.The cultured HRMC(10 6)were intervened with the same concentration of BSA and AGEs-BSA(160,320 and 640μg/ml)for 48h,or 640μg/ml for 1,4,8,24 and 48h.The IL-8 level in supernatant was measured by ELISA.Results The IL-8 concentration began to elevate(71.9±9.2pg/ml,P<0.05)after 4h intervention with AGEs-BSA,and further increased(546.6±49.7,652.1±42.8 and 2 485.2±99.9pg/ml,P<0.01)when HRMC incubated with AGEs-BSA for prolonged time(8?24 and 48h).The level for IL-8 also increased (178.1±15.6,475.4±18.9 and 2 485.2±99.9pg/ml,P<0.01)with the increase of AGEs-BSA concentration(160,320 and 640μg/ml).Conclusion The AGEs-BSA can stimulate the secretion of IL-8 in cultured HRMC in concentration and time dependently.
