2种不同ING1 mRNA剪接体在大肠癌组织中的表达研究 被引量:3

摘要 目的 探讨抑癌基因ING 1在大肠癌发生和进展中的作用和意义。方法 用逆转录 聚合酶链反应 (RT PCR )检测ING 1mRNA的表达 ;用单链构像多态性分析 (PCR SSCP)方法进行突变检测 ;用微卫星位点进行杂合性缺失 (LOH )分析。结果p3 3 /ING 1mRNA和p47/ING 1mRNA在大肠癌组织中的表达强度明显弱于正常大肠黏膜组织者 ;这 2种不同ING1mRNA剪接体之间的表达强度相一致。DukesC、D期患者癌组织中这 2种ING 1mRNA的表达强度明显弱于DukesA、B期者。 3 5例中均未发现ING 1基因突变 ,仅有 4例 ( 11.4% )出现微卫星位点的LOH。结论 p3 3 /ING1mRNA和p47/ING 1mRNA在大肠癌组织中的表达强度相一致 ;它们的低表达与大肠癌的发生、进展有密切关系。ING 1基因的突变和杂合性缺失少见 。 Objective To study the effect and significance of tumor suppressor gene ING1 in the genesis and progression of colorectal carcinoma.Methods ING1 mRNA expression was examined by RT-PCR.Mutation analysis was detected by PCR-SSCP.LOH analysis was undertaken by using microsatellite markers.Results The expression intensities of p33/ING1 mRNA and p47/ING1 mRNA in the colorectal cancerous tissues are significantly weaker than those in the normal ones.The expression intensities between these two ING1 mRNA splices are coincident.The Expression intensities of these two ING1 mRNA splices in the colorectal cancerous tissues of the patients in Dukes C and D are significantly weaker than those in Dukes A and B.No mutation was detected in 35 patients.LOH was only founded in 4 cases(11.4%).Conclusion The expression intensities between these two ING1 mRNA splices are coincident;their low-expression correlate significantly with the genesis and progression of the colorectal carcinomas.There is few mutation and LOH of ING1 gene,its decreased expression maybe happen in the stage of transcription or post-transcription.
