摘要
目的 :观察不同剂量 X射线对小鼠腹腔巨噬细胞转录因子 NF- κB的影响。方法 :用凝胶电泳迁移率变化分析法得到电泳带 ,用 Phosphor Im ager扫描仪分析数据。结果 :不同剂量 X射线照射后小鼠腹腔巨噬细胞NF- κB异源二聚体 p6 5 / 5 0的 DNA结合活性与假照组相比有所增强 ,在 0 .10 0 Gy处出现峰值 ,然后逐渐回降 ,到6 .0 0 0 Gy时再一次升高 ;但同源二聚体 p5 0 / 5 0的 DNA结合活性无明显变化。结论 :高、低剂量 X射线诱导小鼠腹腔巨噬细胞转录因子 NF- κB的 DNA结合活性增强以异源二聚体 p6 5 / 5 0增强为主。
Objective To study the effect of different doses of X-irradiation on transcription factor NF-κB in peritoneal macrophages of mice. Methods Electrophoresis mobility shift assay (EMSA) was used to separate the binding bands of NF-κB with DNA probe, the results were analyzed by Phosphor Imager Scanning System. Results The DNA binding activity of NF-κB heterodimer p65/50 in peritoneal macrophages of irradiated mice increased compared with that of sham-irradiation mice, peaked after irradiation with 0.100 Gy of X-rays, followed by gradually returned to the sham-irradiation level, then increased again after irradiation with 6.000 Gy of X-rays; but the DNA binding activity of NF-κB homodimer p50/50 had no changes after irradiation. Conclusion Irradiation with both low dose and high dose of X-rays can induce the increase of the DNA binding activity of NF-κB predominantly heterodimer p65/50 in peritoneal macrophages in irradiated mice.
