大鼠舌下神经损伤后舌下神经核内星形胶质细胞和凋亡神经元的关系

Relationship between astrocytes and apoptotic neurons in hypoglossal nuclei after hypoglossal nerve injury in rats

摘要 目的:研究大鼠舌下神经损伤后舌下神经核内星形胶质细胞和运动神经元的反应及其相互关系,并探讨其可能机制。方法:84只大鼠随机分为2组:舌下神经切断组和舌下神经假切断组。各组再随机分为1,3,7,15,21,30和60d共7个时间组。采用Tunnel法和免疫组织化学ABC法,观察舌下神经切断后舌下神经核内神经元的凋亡和星形胶质细胞胶质原纤维酸性蛋白(GFAP)的变化进行了研究。结果:舌下神经横断伤后第3天舌下神经元出现凋亡,15d后达凋亡高峰,与假手术对照组相差显著(P<0.01);舌下神经横断伤可引起手术组同侧舌下神经核内GFAP阳性产物明显增加,与未损伤对侧相比差异有显著性意义(P<0.01)。GFAP增加在伤后第1天出现,为(23.48±4.89)个/mm2,15d(32.68±5.14)个/mm2和21d(33.03±4.37)个/mm2达到高峰,突起增多、粗短,胞体变大,分布于整个舌下神经核内;双重标记显示凋亡细胞周围为GFAP阳性星形胶质细胞包绕。结论:舌下神经横断伤后舌下神经核内星形胶质细胞可能在舌下神经元凋亡过程中发挥重要作用。 AIM:To study the response of astrocytes and motoneurons in hypoglossal nuclei and their interrelation after hypoglossal nerve injury in rats,and explore the possible mechanism. METHODS:Eighty four male rats were randomly assigned into two groups:hypoglossal nerve transection group and sham operation group,and then each group was subdivided into seven time groups of 1,3,7,15,21,30 and 60 days. Tunnel method and immunohistomchemical ABC technique were used to determine the changes of glial fibrillary acidic protein(GFAP) like astrocytes and apoptosis of motorneurons in hypoglossal nuclei following the unilateral hypoglossal nerve transection. RESULTS:The hypoglossal neurons apoptosis was firstly detected 3 days post axotomy,and reached the apoptotic peak of hypoglossal neurons at 15 days in axotomy rats,significantly different from that in the sham operation rats(P< 0.01).Axotomy provoked a significant increase of GFAP immunoreaction products within the axotomized hypoglossal nuclei as compared to the contralateral uninjured nuclei(P< 0.01).The enhanced GFAP was firstly observed 1 day post axotomy[(23.48±4.89) per mm2],and the maximal GFAP staining occurred at 15 days[(32.68±5.14) per mm2] and 21 days[(33.03±4.37) per mm2] following axotomy.Branched,fat and short processes and augmented cell bodies distributed throughout the hypoglossal nuclei.Double immunohistochemical staining showed that the apoptosis neurons were closely surrounded by GFAP positive astrocytes. CONCLUSION:Astrocytes may play a key role in the apoptosis of hypoglossal neurons after the hypoglossal nerve transection.
