电泳隐秘变异的测定及其遗传变异增量的估计

摘要 电泳技术揭示了生物中大量的遗传变异。但一般的常规电泳并不能测出所有的遗传变异。利用连续电泳、热变性、尿素变性等技术可以检测出自然群体中更多的电泳隐秘的蛋白质变异。然而,最有效的技术是肽谱分析。利用此方法可以最大限度地测出群体中某一基因位点的变异程度。本文综述了各种特殊的电泳技术及其应用分析的实例,介绍了美国著名遗传学家F.J。Ayala 探讨电泳隐秘变异导致的遗传变异增量的作用所建立的估算模型,由此对蛋白质变导的“电荷态模型”提出了异议。 Electrophoretic techniques have revealed a great amount ofgenetic variation in living thingos.But conventional electro phoresis cannotdetect all of the genetic variatection.A variety of technique—includingsequential electrophoresis and heat or urea denaturation—have been usedto detect more electrophoretically cryptic protein variation in naturalpopulations.The most effective technique,however,is peptide mapping,using which can detect the variation of a locus to the utmost extent.Thisarticle summarizes various methods of electrophoretic techniques and theirapplications,introduces Ayala,a famous American genetist,advanced amodel in order to explore the effect of cryptic variation on currentestimates of genetic variation,and showed that the existing evidences wereinoconsistent with the charge-state model.
