β_1整合素及纤维粘连蛋白与肺癌细胞凋亡关系的实验研究 被引量:3

摘要 目的 研究 β1整合素及纤维粘连蛋白与人肺癌细胞凋亡之间的关系。方法 体外无血清培养条件下 ,以培养在包被纤维粘连蛋白底物上的人肺腺癌SPC -A - 1细胞为靶细胞 ,用不同浓度抗 β1整合素单克隆抗体阻断β1整合素与纤维粘连蛋白相互作用 ,采用苏木素 -伊红 (HE)染色 ,吖啶橙荧光染色 ,DNA琼脂糖凝胶电泳检测细胞凋亡情况 ,并计算凋亡指数。结果 抗 β1整合素单克隆抗体对SPC -A - 1细胞凋亡有明显的促进作用 ,在一定的范围内 ,呈时间、剂量依赖性。结论 肺癌细胞凋亡与 β1整合素受体有密切关系 ,且 β1整合素受体与其特异配体纤维粘连蛋白的相互作用能促进细胞生存 。 Objective To study the relationship among β 1 integrin,fibronectin and lung cancer cell apoptosis in vitro.Methods Human lung adenocarcinoma SPC-A-1 cell line cultured on the peri dishes coated by fibronectin in serum-free medium in vitro was treated with monoclonal antibody against β 1 subunit at different concentrations to block the interaction between β 1 integrin and fibronectin.HE staining, acridine orange(A0)staining and DNA electrophoresis assay were used to assay cell apoptosis.Results SPC-A-1 cell apoptosis was induced significantly by monoclonal antibody against β 1 subunit. Within some limits,the effect was dose- and time-dependent.Conclusions Lung carcinoma cell apoptosis may be related to β 1 integrin. Cell survival was increased by the interaction between β 1 integrin and fibronectin and cell apoptosis was induced by blocking the interaction.
