谷氨酸生产菌S_(9114)中谷氨酸脱氢酶分离纯化条件的初步研究 被引量:1

摘要 ;为获得谷氨酸脱氢酶(g1utamatedehydrogenase,GDH)分离纯化的较好效果,文章对Frenchpres surecellpress的菌体破碎条件和DEAE-纤素柱分离条件进行了初研究。结果表明:细胞破碎的较好条件为Frenchpressurecellpress18000pa破碎两次;DEAE-纤维素柱层析GDH的较好条件为柱洗脱线性梯度盐浓度范围为0.2~0.6mo1/L,上样量为5~8mL,柱流速为2~3mL/min。 In order to require the optimal purification condition of glutamate dehydrogenase fron Corynebacteriumglutacium S_(9114) on DEAE-cellulose column were investigated.It was shown that the cells were disrupted twice using a French pressure cell press at 18000Pa;the chromatographic separation of DEAE-cellulose column were performed with a linear gradient elution of NaCl from 0.2 to 0.6mol/L in 0.025mol/L Tris-HCl buffer(pH7.5),with the extracts of 5~8mL injected and at flow-rate of 2~3 mL/min,respectively.
