摘要
通过对12个氨基酸的4-氯-7-硝基苯-氧二氮杂茂(NBD-Cl)衍生物在使用同一个微乳液缓冲液和相同的仪器操作参数下连续27次运行的分析,考察了其迁移窗口、迁移顺序以及部分衍生物的相对标准偏差(n=6)、分辨率以及理论塔板数的变化,并对影响运行重现性的原因进行了分析,归纳为3个主要方面:温度的影响,微乳液本身形成和稳定过程的影响和毛细管壁变化的影响。
Run-to-run reproducibility in microemulsion electrokinetic chromatography was investigated and tested with separation of twelve amino acids by microemulsion electrokinetic chromatography combined with laser-induced fluorescence detection after derivatization with 4-chloro-7-nitrobenzo-2-oxa-1, 3-diazol(NBDCl). In a batch of 27 runs using the same microemulsion buffer and same operation parameters, theoretical plates, resolutions, RSD(n=6), migration window and order were discovered to be changing and their causes were also analyzed.
