抗棘球蚴人源单链可变区抗体抑制人体包虫原头节的初步实验

摘要 目的 建立人体包虫原头节的体外培养模型 ,评价抗棘球蚴人源单链可变区抗体(ScFv)抑制人体包虫原头节的效果。方法 无菌抽取人体包虫囊肿原头节 ,与ScFv高 (1mg/L)、中(0 .1mg/L)、低 (0 .0 1mg/L)剂量组和对照组 (等量的培养液 )共同培养 ,比较各组原头节死亡率和残余ScFv浓度 ,观察原头节形态学改变。结果 高剂量ScFv组原头节死亡率与对照组的差异均有统计学意义 (P <0 .0 1) ,但中、低剂量组与对照组死亡率的差异无统计学意义 (P >0 .0 5 ) ,随作用天数增加 ,高剂量ScFv组原头节结构逐渐被破坏、皱缩、崩解。结论 该ScFv具备体外抑制人体原头节的作用 ,其抑制作用与剂量和时间有关。 Objective To set up an in vitro model of h um an echinococcosis protoscolex and evaluate the anti-protoscolex effect of ScFv. Methods Protoscolex were extracted from surgical samples , then mixed with high(1mg/L), middle(0.1mg/L), low (0.01mg/L)doses ScFv or sali ne without ScFv as control. Morphological changes, mortality of protoscolex and ScFv density in 4 groups were observed at different times. Results There was statistical significance between high does ScFv group and c ontrol group, while the middle and low groups failed to show significant inhabit ation. Protoscolex mixed with high dose ScFv became distorted and lost its norma l construction and finally lysed. Conclusion Anti-echino coccosis ScFv can inhibit protoscolex in vitro, and its effect was positivel y related to the dosage and incubation time. [
