摘要 目的 观察血清剥夺致PC12细胞死亡及对细胞周期的影响。方法 流式细胞术检测在含 5 %和 2 %胎牛血清的培养基中 ,PC12细胞死亡和细胞周期的改变。结果 随着血清浓度的下降 ,PC12细胞死亡或凋亡增加 ,同时 ,S期的细胞所占百分比降低 ,G2 期细胞所占的比例增加 ,在含 2 %胎牛血清的培养基中培养的细胞更加明显。结论 ①PC12细胞经血清剥夺培养后产生的损伤可能引发了细胞周期阻滞 ,当损伤未得以及时修复时 ,导致细胞凋亡。②在细胞周期和细胞死亡之间可能存在密切的联系。 Objective To observe the changes of cell death and cell cycle of PC12 cells in serum-deprivation medium. Methods Flow cytometry was used to detect the changes of cell death and cell cycle of PC12 cells cultured in medium containing 5 % and 2 % serum. Results As the concentrations of serum were decreased, the percentage of dead or apoptotic cells was increased. At the same time, there was a decline in the percentage of cells in S phase and an increase in the percentage of cells in G_2. Such changes of cell cycle were more obvious in the cells cultured in medium containing 2 % serum. Conclusion The damage of PC12 cells induced by the serum deprivation led to cell cycle arrest, even the apoptosis when the damage couldn't be repaired on time. There may be a close relationship between cell cycle and cell death.
