摘要
目的 分离和纯化母婴血型不合母体的特异性高效价的IgG抗体。 方法 采用半饱和硫酸铵低温沉淀法分离IgG抗体 ,SephedexA5 0层析柱纯化 ,然后用B型红细胞 (RBC)吸收放散IgG ,用聚乙二醇 (PEG)沉淀 ,真空泵脱水回收IgG。结果 2 0 0ml血浆经半饱和硫酸铵低温沉淀后得IgG粗制品 2 2 6 g ,SephedexA5 0层析柱纯化后得纯制品 1 84g ,经B型RBC吸收放散后得特异性IgG 0 5 5 g ,回收率达 2 9 89% (0 5 5 / 1 84 )。 结论 采用半饱和硫酸铵低温沉淀法分离高效价IgG抗体 ,可以获得高纯度的IgG。
Objective To isolate and purify the high titer specific IgG antibody in fetometernal blood type incompatible mother. Methods High titer specific IgG antibody was precipitated by using saturated ammonium sulphate, purified by sephedex A50 column, and B type red blood cell was used for IgG sorption-elution, the purified IgG was concentrated by polyethylene glycol and lyophilized. Results 2.26 g crude product was got from 200 ml after sedimentation of saturated ammonium sulphate, and 1.84 g was left after sephedex A50 chromatography. 0.55 g specific IgG antibody was received finally after red blood cell sorption-elution with the recovery ratio of 29.89 % (0.55/1.84). Conclusion High purity and efficient isolated specific IgG could be obtained by using saturated ammonium sulphate under low temperature.
