摘要 目的 用RNA干扰技术特异性抑制SARS冠状病毒在非洲绿猴肾细胞VeroE6中的增殖。方法 体外转录合成针对SARS CoVRNA的siRNA ,使用脂质体转染的方法导入细胞 ,观察并估算病毒感染细胞病变值 ,MTT法检测细胞存活率 ,RT PCR确定胞内病毒RNA水平。结果 成功筛选出 4组siRNA能够显著降低SARS CoV感染细胞中病毒RNA水平 ,提高感染细胞存活率。结论 应用RNAi技术可以有效阻止病毒在细胞中增殖 ,为治疗病毒性疾病提供了新思路。 Objective To specifically inhibit SARS-CoV proliferation in African green monkey kidney cell line Vero E6 with RNAi technology.Methods siRNAs targeting SARS-CoV RNA were obtained by in vitro transcription and transfected into cells with lipofectin. We observed and evaluated the CPE (cytopathic effect), the survival of the infected cells with MTT assay as well as the viral RNA level in the infected cells.Results Four siRNAs which could dramatically knock-down SARS-CoV RNA in the infected cells and enhance the survival of the cells were obtained.Conclusion siRNAs can effectively inhibit SARS-CoV proliferation in infected cells.
