摘要
目的 了解应用绒毛细胞进行D3S135 8、D13S317、vWA、D2 1S11、D18S5 1、FGA、D5S818、D7S82 0、D8S1179等9个STR基因座在汉族胎儿的遗传多态性分布及其在法医学和产前基因诊断的应用价值。方法 采用复合荧光标记扩增技术 ,用绒毛细胞和部分蜕膜细胞对上述 9个STR基因座进行分型 ;扩增产物用ABI - 310DNA序列分析仪电泳检测 ;数据分析统计采用ABI-GeneScan3.1、ABI-Genotype4 .0软件和 χ2 检验。结果 ( 1)所有绒毛和蜕膜样本复合扩增成功 ,没有相互间的污染。 ( 2 )获得了绒毛细胞上述 9个STR基因座的基因频率分布资料。 ( 3)绒毛细胞和外周血细胞的 9个STR基因座的等位基因频率无显著性差异 (P >0 .0 5 )。结论 用绒毛细胞进行上述 9个STR基因座的多态性分布研究是可行的 。
Objective: To have a thorough knowledge of the genetic polymorphism of 9 STR loci in Han's fetus by CVS and its application in forensic science and prenatal gene diagnosis. Methods: Using multiplex fluorescent amplification technique, 9 STR loci in 102 chorionic villus samples and 30 deciduous samples of Han's fetus were amplified, the PCR products were lectrophresised by ABI 310 sequencer. The results were analysed by ABI-GeneScan3.1、ABI-Genotype4.0 software andχ 2 test. Results: (1)All samples were amplified successfully. The 9 STR loci of CVS correspond to deciduous samples. (2)The polymorphism of 9 STR loci have been obtained in Han's fetus by CVS. (3)The frequencies of 9 STR loci were not statically significient between chorionic villus samples and peripheral blood samples.Conclusion Study on genetic polymorphism of 9 STR loci in Tianjin Han by CVS is available.The data which we obtained from CVS can be used not only in genetic research and population investigation,but also in forensic identification and prenatal paterinity.
