Effect of homocysteine on proliferation of vascular smooth muscle cells

摘要 目的 :探讨同型半胱氨酸 (Hcy)对血管平滑肌细胞 (VSMC)生长的影响。方法 :采用MTT法检测细胞数量及流式细胞术测定细胞周期。结果 :5× 10 -4mol/LHcy干预处于生长静止期的细胞 4 8、72h细胞数量较对照组明显增加 (P <0 .0 1) ,分别增加了 5 0 .75 %和 84 .83%。当采用 1× 10 -4,2 .5× 10 -4,5× 10 -4,1× 10 -3mol/L的Hcy干预细胞 72h后 ,其细胞数量分别较对照组增加了4 1.5 6 % ,5 8.4 4 % ,6 6 .2 3% ,74 .0 2 %。并且当 5× 10 -4mol/LHcy干预细胞 2 4、36、4 8h后 ,其S期、G2 /M期在细胞周期的比例也明显增加 (P <0 .0 1)。结论 :Hcy能明显诱导VSMC增殖 ,并呈现时间和剂量依赖关系 ,提示Hcy通过诱导VSMC的增殖可导致动脉粥样硬化的发生。 Objective:To explore the effect of homocysteine(Hcy) on proliferation of vascular smooth muscle cells(VSMC). Method: MTT to test cell number and flow cytometer to determine the cell cycle. Result:The cell number increased distinctly when quiescent cells were exposed to 5×10 -4 mol/L Hcy for 48 and 72 h (P< 0.01). The cell number increased 50.75% and 84.83% compared with control group. Exposed cells to 1×10 -4, 2.5×10 -4,5×10 -4,1×10 -3 mol/L Hcy for 72 h respectively, we observed that the cell number increased 41.56%, 58.44%, 66.23%, 74.02% compared with control group. The S phase and G 2/M phase were also increased when cells were treated Hcy for 24 h, 36 h, 48 h (P< 0.01). Conclusion:Hcy can promote the proliferation of VSMC obviously and the prolifertion effects are in a manner of dose-dependent and time-dependent. The result suggest that Hcy induce the atherosclerosis by the way of facilitating the proliferation of VSMC.
