摘要 目的 探讨2-(3-羧基-1-丙酰氨基)-2-脱氧-D-葡萄糖是否具有诱导人食管癌细胞Eca-109发生凋亡的作用。方法 采用体外细胞培养实验力法,应用MTT法、流式细胞仪检测、DNA琼脂糖凝胶电泳、透射电镜等技术,观察药物对细胞生长的抑制率及细胞的形态学变化,检测凋亡峰及DNA Ladder。结果 MTT法测得细胞抑制率具有显著的时间及浓度依赖性;透射电镜下可见到凋亡典型的形态学变化,流式细胞仪检测显示细胞周期的G1期阻滞及凋亡峰;DNA琼脂糖凝胶电泳显示清晰的DNA梯状电泳。结论 2-(3-羧基-1-丙酰氨基)-2-脱氧-D-葡萄糖能够明显的抑制人食管癌细胞Eca-109的增殖,并诱导其发生凋亡。 Objective To investigate whether 2- (3- carboxy- 1 - ocoprolyl) amino - 2 - deoxy - D - Glucose could induce apoptosis in human esophageal cancer cell line Eca -109. Methods In vitro experiments, MTT assay, Transmission electron microscope, DNA agarose gelelectriphoresis, Flow cytometry were performed to determine the cell growth inhibitory rate and observe cell morphological change, cell proliferation cycle and apoptosis index. Results 2 - (3 - carboxy - 1 - oxoprolyl) amino - 2 - deoxy - D - Glucose inhibited the growth of esophageal cancer cell line Eca- 109 in a time-and-dose-dependent manner, typical apoptotic morphological change were observed. Flow cytometry analysis showed that an arrestment at G1 phase, an apoptosis peak presented before sub - G1 cell peak. A mark DNA Ladder bands was formed in electrophoresis. Conclusion 2 - (3 - carboxy - 1 - oxoprolyl) amino - 2 - deoxy - D - Glucose could inhibit the proliferation and obviously induce apoptosis in human esophageal cancer cell line Eca - 109
