bFGF,GDNF,PDGF,IGF-I在大鼠脊髓中的定位表达 被引量:2

The localization and expression of bFGF,GDNF,PDGF,and IGF-I on the spinal cord of rat.

摘要 目的 碱性成纤维细胞生长因子 (bFGF)、胶质细胞源性神经营养因子 (GDNF)、血小板源性生长因子 (PDGF)和胰岛素样生长因子 -I (IGF -I)对神经细胞起特殊的营养作用 ,但它们在脊髓中的定位分布和生物学作用还不十分清楚。本研究旨在探索这一问题 ,方法 取正常成年SD大鼠脊髓分别用抗bFGF ,GDNF ,PDGF ,IGF -I抗体行免疫组化染色 ,观察上述 4种神经营养因子免疫阳性反应物在大鼠脊髓中的分布。结果 bFGF ,GDNF ,PDGF ,IGF -I的免疫阳性反应物主要见于脊髓灰质内各板层神经元 ,腹角和背角深部明显 ,背角浅层较少见。但II板层还可见bFGF阳性细胞和GDNF阳性膨体 ,中央管可见PDGF、bFGF阳性的室管膜细胞。结论 bFGF ,GDNF ,PDGF ,IGF Objective bFGF,GDNF,PDGF and IGF-I were designated as crucial neurotropic factors which were identified in recent years.Several evidences had demonstrated that these factors might be a nutrient material of neurons. However,their biological action and the localization in the spinal cord had been unclear.The aim of the present study was to elucidate this problem.Methods Spinal cords of adult rat were stained using bFGF,GDNF,PDGF,IGF-I antibody by immunohistochemistry method.Results The immunohistochemical positive products on bFGF,GDNF,PDGF and IGF-I were observed in the neurons of gray matter,especially in both ventral horns and the deep of dorsal horns.The bFGF immunohistochemical positive neurons and GDNF positive varicosities were also present in spinal laminal II.Both PDGF and bFGF positive ependyma cells were observed around central tube.Conclusion bFGF,GDNF,PDGF,and IGF-I play an important role in maintaining the biological function of neurons in the spinal cord.
